Darrelle attended the University of Pittsburgh and played football for the Panthers. In his sophomore year, 2005, he was named first-team All-Big East at cornerback. He skipped his senior year of college to enter the 2007 NFL Draft. He began his rookie season with the New York Jets, where he was coached by veteran Kerry Rhoades.

He established himself as a starter in his rookie season, but despite stellar performances, the Jets’ defense couldn’t keep up, ranking 28th and 29th in his first two seasons, with teams not testing his side of the field. In 2009, he was named to the Pro Bowl.

He guided the Jets to their first AFC Championship game since 1998, but they were defeated. He continued to play an important role in many of the Jets’ games, putting in some outstanding performances. He was later traded to the Tampa Bay Buccaneers. The Kansas City Chiefs signed him to a two-year, $11 million contract. He was released after only one year and six games. In 2018, he announced his retirement, and the Jets held a mini-commemoration ceremony and created a makeshift Revis Island in his honor.
